She is known for her performances in the original West End production of Spamalot and the 2010 Regent's Park revival of Into the Woods as well as The Wizard of Oz (as the Wicked Witch of the West). Her work has earned three Olivier Award nominations. She was cast as Septa Unella in Game of Thrones' fifth season in 2015. In 2015, she was also cast as an ensemble actor in Les Miserables' 2012 adaptation and Winter Ridge as a primary character. Waddingham was nominated for a Golden Globe Award. She has been playing the character of Rebecca Welton on the Apple TV+ comedy series Ted Lasso from 2020.
